Antibody technology and phenotypic discovery

Therapeutic antibodies are the fastest growing class of drugs. Typically antibodies are developed against predefined target molecules. As a consequence target space is rather limited. Phenotypic drug discovery, instead focuses on development of drugs with a specific function without prior knowledge of the target of the drugs. This thesis describes and discusses the development of antibodies against the large range of targets displayed on cells using phage display technology, and defines and discusses approaches to identify binders and their targets in the complex mixture of antibodies that result from such a process.
